I'm using Canon EOS 60D. According to Canon's site, it is capable of all SD, SDHC and SDXC cards. I have had emailed SanDisk technical support regarding this issue. And the answer I got is that the new Extreme® Pro™ SDHC™ UHS-1 card is capable of supporting both FAT32 and exFAT.

But, what happen if I put the UHS-I card in a camera that does not support or optimized to UHS-I, will get the 45MB/s or will function as a Class-10 card at 200X (30MB/s)? We have been advised that SanDisk are going to discontinue the former SDHC extreme Class-10 cards (30MB/s version) for the new UHS-I.

Is the camera compatible with SDHC/SDXC and UHS-1 Memory Cards?
https://integralmemory.com/faq/what-uhs-1
"UHS-1" or more accurately "UHS-I" stands for "Ultra High Speed" - 1 and is a speed class for SDHC and SXDC memory cards. UHS-I has a bus interface speed of up to 104 MB/s. An SDHC UHS-I card will work in any SDHC compatible device at lower speeds, but to take advantage of the UHS-I speed, a UHS-I compatible device is essential.
